Humanwild-typep53proteiniscomposedof393aminoacidresidueswithseveraldistinctregions.TheN-terminalactivationdomainallowsp53proteintorecruitthebasaltranscriptionmachineryandactivatetheexpressionoftargetgenes,whereasthecoredomainbindstotargetDNAinasequence-specificmannerandthemajorityofmutationsfoundinhumantumorsoccurintheregionofthegeneencodingthisdomain(1-3).TheC-terminaldomainiscomposedofpredominantlybasicresiduesandmodificationoftheC-terminalbasicdomain,includingacetylation,glycosylationandphosphorylation,isanessentialmechanismforregulatingp53function(4-6). |
